L-Tyrosine is an amino acid that makes up protein. It has ionized aromatic side chain and is hydrophilic. Tyrosine is produced by hydroxylation of phenylalanine in human and animal body. Therefore, when phenylalanine nutrition is sufficient, it is an unnecessary amino acid.
The catabolism of tyrosine is first catalyzed by intrahepatic tyrosine transaminase, which needs pyridoxal phosphate as coenzyme. P-hydroxyphenylpyruvate, through the action of p-hydroxyphenylpyruvate hydroxylase, causes the oxidative decarboxylation and transfer of side chain pyruvate, and the hydroxylation of the ortho position of benzene ring, thus forming urinary black acid (dihydroxyphenylacetic acid). The enzyme is a copper containing metalloprotein, which needs ascorbic acid as coenzyme and consumes molecular oxygen. Under the catalysis of uromelanic acid dioxygenase (uromelanic acid oxidase), uromelanic acid splits the benzene ring to produce maleic diacetyl acetic acid; the enzyme is a metal protein containing iron, and the reaction requires the participation of a molecule of oxygen. Glutathione is needed as coenzyme for the conversion of maleic diacetate to fumarate via the action of corresponding isomerase. At last, it is hydrolyzed to fumaric acid and acetoacetate by corresponding hydrolase, so tyrosine is not only glycogen but also ketogenic amino acid.
