        Tyrosine iodization refers to the iodide formed by replacing the third and fifth protons on the tyrosine benzene ring with activated iodine. The process of iodization was mainly carried out on thyroglobulin molecules near the top cells in the acinar cavity.
    The steps of tyrosine iodization and synthesis of T4 and T3 are as follows: tyrosine reacts with iodine to form monoiodide tyrosine (MIT) and diiodine tyrosine (DIT), and two molecules of DIT are connected to form T4. T3 is mainly produced by the deiodination of T4 in the surrounding tissues, but it can also be produced by the connection of 1-molecule MIT and 1-molecule DIT in the epithelial cells.
    In epithelial cells, the above-mentioned iodization reaction is carried out on the tyrosine residues of thyroglobulin molecules, not on the free tyrosine. Therefore, the synthesis of thyroid hormone is carried out on the thyroglobulin molecule. Thyroglobulin is a kind of glycoprotein. Thyroglobulin in human body is composed of four polypeptides. Each polypeptide chain has a molecular weight of 160000. The polymerization of four polypeptides has two forms: one is the disulfide bond connection between four polypeptides; the other is the disulfide bond connection between two polypeptides to form a dimer, and then the covalent bond connection. The stereoscopic structure of the complete tetramer of thyroglobulin molecule must be complete in order to make some tyrosine residues have proper spatial position, so as to facilitate the iodization and the binding of the iodized tyrosine to th. There are about 120 tyrosine residues per molecule of thyroglobulin, only a few of which are iodized. It was found that MIT and T3 in thyroglobulin molecules increased relatively when iodine deficiency occurred in rats, which may be a compensatory effect to deal with iodine deficiency. The iodine needed for the synthesis of T3 is 1 / 4 less than T4, and the activity of T3 is higher than T4. Because the ratio of T4 / T3 in thyroglobulin molecule is 12, while the ratio of T4 / T3 secretion per day is 3. It can be seen that T4 can be transformed into T3 in the thyroid.
